Isolation and Culture of Cotton Protoplasts

ABSTRACT

Protoplasts of Gossypium harknessii and G. hirsutum were enzymatically isolated from callus cultures. Purification was accomplished by floating protoplasts over a sucrose solution. Salt composition of the culture media that were tested affected protoplast growth. Protoplasts were grown in culture to cell colonies.
